Antithrombin (AT) is a plasma protein that acts, in the presence of heparin, to covalently inhibit several serine proteases (e.g. thrombin-IIa and factor Xa) involved in blood coagulation. VAT-10 is a purified AT product designed for use in laboratory inhibition assays (Anti-IIa and Anti-Xa tests) of heparin and LMW-heparin.

Each vial of VAT-10 contains 10 IU of Human Antithrombin, co-lyophilized with Tris buffer and mannitol stabilizers. Sealed glass container has a 20 ml capacity for reconstitution.

Human plasma origin: negative tests for viral pathogens
Synonym: AT, AT III

Purity: Single band by SDS-PAGE analysis
Molecular weight: ~58,000 Da
Activity per vial: ~10 IU/vial vs International Reference Standard
Activity of source enzyme: > 6 IU/mg

Product should be stored refrigerated (2-8°C) and is stable until expiry in the unopened state. Stable 1 month at 2-8°C once reconstituted. Shipment of unopened product does not require refrigeration.

AT binds to heparin of LMW-heparin to form a 1:1 complex (AT-hep), which subsequently inactivates either thrombin (IIA) or factor Xa. In linked enzyme inhibition assays (i.e. Anti-IIa and Anti-Xa tests) increasing the heparin to AT ratio results in a proportional decrease of enzyme activity (IIa or Xa) measured chromogenically.
